The eggplant (Solanum melongena L.) belongs to the Solanaceae family and is an important plant species in the world. In vitro production of haploid/doubled haploid plants from microspores is a useful technique in plant breeding to produce commercial cultivars. This research aimed to investigate plant regeneration from the calluses obtained by isolated microspore culture in eggplant. This experiment was done using MS medium. First, the calli larger than ۲ mm, produced through microspore culture, were selected and cultured in MS medium containing ۲ mg.l-۱ BAP, ۰.۲ mg.l-۱ NAA, ۰.۲۵% phytagel and ۳% sucrose for callus proliferation. After one month, a suitable volume of callus was produced. Then, these calli were transferred to MS medium containing ۲ mg.l-۱ kinetin, ۱ mg.l-۱ BAP, ۰.۶۵% agar-agar, and ۳% sucrose for shoot-inducing. After two months, shoot regeneration and formation of green meristem points in the calli were checked. On average, ۷ normal shoots and ۲۵ meristem points were observed in each explant (a piece of callus with a diameter of about ۱ cm). Shoots were separated and the calli containing meristem points were transferred to MS medium containing ۰.۵ mg.l-۱ kinetin. After one month, most meristem points grew and produced new shoots. All regenerated shoots were transferred to a hormone-free MS medium containing ۱۰۰ mg.l-۱ casein hydrolyzate and ۳% sucrose for rooting. The concentrations of vitamins in this culture medium had been doubled. After one month, the rooting was done successfully. The obtained plantlets were transferred to the pots containing a mixture of perlite and peat moss (۱:۱) and their acclimatization process was performed. This plant regeneration protocol had good efficiency in the studied cultivar and can be used in breeding programs.
