چکیده مقاله:

The most common form of cancer in women and the leadingcause of death, breast cancer is caused by a variety of mechanisms,including changes in gene expression and function. Byinhibiting the expression of antiapoptotic genes and activatingcaspases, curcumin decreases proliferation and induces apoptosisin cancer cells.Our study examines the effects of nanocurcumin on key breastcancer factors by measuring gene expression for the lnc-MEG۳and lnc-FOXCUT genes in MCF-۷ breast cancer cells treatedwith nanocurcumin. To determine the drug concentration effective,MCF۷ cells were treated with ۲۵ mM nanocurcuminafter MTT assay. For analysis of the expression of MEG۳ andlnc-FOXCUT, RNA was extracted, cDNA synthesized and RealTime PCR was performed. To evaluate apoptosis, flow cytometrywas used.The results indicated that nanocurcumin increased the expression of lnc-MEG۳, but decreased the expression of lnc-FOXCUT,and that ۹۱ percent of cells underwent apoptosis after ۴۸hours of treatment. At the above-mentioned dose, nanocorcinomsignificantly increases the expression of lnc-MEG۳, a tumorsuppressor gene, while reducing FOXCUT. DNA methylationchanges may cause these alterations that lead to apoptosis
